目的 探讨二维斑点追踪技术评价牛磺酸对宫内发育迟缓(IUGR)大鼠心功能的影响。 方法 采用限制蛋白饮食方式建立IUGR大鼠模型,IUGR大鼠出生后随机分为IUGR组、IUGR+低剂量牛磺酸组及IUGR+高剂量牛磺酸组,以正常饮食出生后大鼠为正常对照组。出生4周后,运用超声斑点追踪技术检测各组大鼠左室心肌应变情况,HE染色检测大鼠心肌细胞数量及体积,免疫组化检测大鼠心肌I型及III型胶原。 结果 (1)出生时,与正常对照组相比, IUGR组、IUGR+低剂量牛磺酸组、IUGR+高剂量牛磺酸组体重均明显减低(P<0.05)。出生4周后,与正常对照组相比,IUGR组体重无明显改变(P>0.05)。与IUGR组相比,IUGR+低剂量牛磺酸组、IUGR+高剂量牛磺酸组体重均无明显改变(P>0.05)。(2)出生4周后,与正常对照组相比, IUGR组左室局部周向峰值应变显著降低(P<0.05)。与IUGR组相比, IUGR+低剂量牛磺酸组左室局部周向峰值应变显著增高(P<0.05),IUGR+高剂量牛磺酸组左室局部周向峰值应变显著增高(P<0.05)。与IUGR+低剂量牛磺酸组相比,IUGR+高剂量牛磺酸组左室局部周向峰值应变显著增高(P<0.05) 。(3)与正常对照组相比,IUGR组大鼠心肌细胞数量减少、体积增大,I型、III型胶原表达增高。与IUGR组相比,牛磺酸组(低剂量+高剂量组)心肌细胞体积减少,I型及III型胶原表达减低,尤其是高剂量牛磺酸组。结论 牛磺酸可以提高IUGR大鼠左室局部周向峰值应变,抑制I型及III型胶原表达,起到抗心肌纤维化、改善IUGR大鼠心功能作用,尤其以高剂量牛磺酸效果更好。减少、体积增大,I型、III型胶原表达增高。与IUGR组相比,牛磺酸组(低剂量+高剂量组)心肌细胞体积减少,I型及III型胶原表达减低,尤其是高剂量牛磺酸组。 |

Objective To evaluate the effect of taurine on cardiac function in intrauterine growth retardation (IUGR) rats by two-dimensional speckle tracking imaging. Methods The IUGR rats were randomly divided into the IUGR group, the IUGR+ low dose taurine group and the IUGR+ high dose taurine group after birth. Four weeks after birth, the local circumferential peak strain of left ventricular was monitored by speckle tracking, the number and volume of myocardial cells were detected by HE staining, and the myocardial type I and type III collagen were detected by immunohistochemistry. Results (1) At birth, compared with the normal control group, the weight of the IUGR group, the IUGR+ low dose taurine group and the IUGR+ high dose taurine group were significantly reduced (P < 0.05). After 4 weeks of birth, there was no significant change in body weight in the IUGR group compared with the normal control group (P > 0.05). Compared with the IUGR group, there was no significant change in body weight in the IUGR+ low-dose taurine group and the IUGR+ high-dose taurine group (P > 0.05). (2) After 4 weeks of birth,compared with the normal control group, the local circumferential peak strain of left ventricular in the IUGR group was significantly reduced (P < 0.05). Compared with the IUGR group, the local circumferential peak strain of left ventricular in the IUGR+ low-dose taurine group was significantly higher (P < 0.05), and that in the IUGR+ high-dose taurine group was significantly higher (P<0.05). Compared with the IUGR+ low-dose taurine group, the local circumferential peak strain of left ventricular in the IUGR+ high-dose taurine group was significantly higher(P< 0.05). (3) Compared with the normal control group, the number and volume of myocardial cells in the IUGR group decreased, and the expression of type I and type III collagen increased. Compared with the IUGR group, the taurine group (low dose + high dose group) showed decreased myocardial cell volume and decreased collagen type I and III expression, especially in the high dose taurine group. Conclusion Taurine can improve the local circumferential peak strain of left ventricular of IUGR rats, inhibit the expression of type I and type III collagen, play an anti-myocardial fibrosis and improve the cardiac function of IUGR rats, especially the effect of high dose taurine is better. |
