目的 制备肿瘤微环境响应性超声/MRI/光学成像纳米粒(Ce6@TA-Fe),观察其性能、体外多模态成像及光热联合氧气增强光动力治疗体外葡萄膜黑色素瘤(UM)的效果。方法 以Ce6、单宁酸(TA)及FeCl3·6H2O制备Ce6@TA-Fe纳米粒,检测其理化性质,观察其体外多模态显像效果,评估光热联合光动力治疗对UM细胞(C918)的体外杀伤作用。结果 成功制备载Ce6和Fe3+的纳米探针(Ce6@TA-Fe),平均粒径(26.51±5.91)nm,电位(-21.5±6.45)mV。体外成像实验证实,Ce6@TA-Fe浓度越高,成像效果越好。激光照射后Ce6@TA-Fe迅速升温,并产生活性氧。Ce6@TA-Fe与C918细胞孵育并经激光辐照后, C918细胞存活率仅(5.71±2.71)%,提示其具有体外杀伤UM细胞能力。结论 所制备Ce6@TA-Fe纳米粒具备体外多模态显像及在联合治疗中杀伤UM细胞的能力。 |

Objective To prepare tumor microenvironment responsive ultrasound/MRI/optical imaging agent nanoparticles (Ce6@TA-Fe), and to evaluate its in vitro performances of multimodal imaging, as well as in photothermal combining with oxygen-enhanced photodynamic therapy for uveal melanoma (UM). Methods Nanoparticles Ce6@TA-Fe were prepared using Ce6, tannic acid (TA) and FeCl3·6H2O, then its physicochemical properties were assessed, its effect of in vitro multimodal imaging was observed, and its cell killing ability during photothermal combining with photodynamic therapy on in vitro UM cells (C918) was explored. Results Nanoprobe Ce6@TA-Fe containing Ce6 and Fe3+ was successfully prepared, with average particle size of (26.51±5.91)nm and zeta potential of (-21.5±6.45)mV. In vitro imaging experiments demonstrated that the higher the concentration of Ce6@TA-Fe, the better the imaging effect. After laser irradiation, Ce6@TA-Fe heated up rapidly and produced reactive oxygen species. The survival rate of C918 cells was (5.71±2.71)% when Ce6@TA-Fe being cocultured with cells under irradiation, indicating that Ce6@TA-Fe had the ability of killing in vitro UM cells. Conclusion The prepared Ce6@TA-Fe had the ability of multimodal imaging and of killing in vitro UM cells during combining therapy. |
