      目的 制备可识别细胞凋亡的膜联蛋白V(annexin Ⅴ)超声靶向微泡(MB),观察其体外寻靶能力。方法 利用生物素-亲和素系统制备靶向MB,并评价其理化性质及靶向配体结合状况。将人甲状腺未分化癌细胞株传代培养至对数期,随机分为3份,以微波消融其中2份,以1份作为对照,获得凋亡细胞并进行验证。于荧光显微镜下观察3组annexin Ⅴ靶向MB与普通MB:A组,凋亡细胞+普通MB;B组,正常细胞+annexin Ⅴ靶向MB;C组,凋亡细胞+annexin Ⅴ靶向MB。结果 仅凋亡细胞可与annexin Ⅴ靶向MB稳定结合。结论 annexin Ⅴ靶向MB可在体外靶向结合凋亡细胞。

      Objective To prepare annexin Ⅴ ultrasound targeted microbubbles (MB) being able to recognize apoptosis, and to explore the in vitro targeting ability. Methods Annexin Ⅴ were prepared with biotin-avidin system, and the physical and chemical properties and binding status of targeted ligand were evaluated. Human undifferentiated thyroid cancer cell lines were subcultured to the logarithmic phase and randomly divided into three sections, one section was used as a control, and the other two underwent microwave ablation to obtain apoptotic cells for verification. Three groups of annexin Ⅴ targeted MB and normal MB were observed under fluorescence microscope, group A consisted of apoptotic cells and normal MB, group B consisted of normal cells and annexin Ⅴ targeting MB, group C consisted of apoptotic cells and annexin Ⅴ targeting MB. Results Only apoptotic cells were stably bounded to annexin Ⅴ targeting MB. Conclusion Annexin Ⅴ targeting MB could target apoptotic cells in vitro.
