目的 评估超声成像技术动态检测大鼠腿部皮下及肌肉间丝素蛋白在体降解及血管化的可行性。方法 制备8%W/V的丝素蛋白溶液,采用超声波振荡制备丝素蛋白凝胶。将40只雄性Wistar大鼠随机分为皮下植入组(n=20)及肌肉植入组(n=20),取0.2 ml丝素蛋白凝胶分别植入大鼠腿部皮下或肌肉间(股直肌与半腱肌的肌间隙)。于植入后的即刻及1、4、8、12、16、18周分别对丝素蛋白凝胶进行二维超声及CEUS,并测量丝素蛋白凝胶的径线值及声学强度;每次成像后,每组随机选取两只大鼠处死,将植入的丝素蛋白凝胶取出进行病理学检查。结果 二维超声测量显示12周后两组的凝胶开始变薄,12周时横径与植入后即刻的凝胶横径差异有统计学意义(P均<0.05);16周时两组凝胶的横径、宽径和纵径与植入时比较差异有统计学意义(P均<0.05)。CEUS示1~8周凝胶内的声学强度呈现相对较低水平(<5 dB),12周后凝胶内部逐渐有造影剂的灌注,16周时声学强度增强可达20 dB,18周时原植入凝胶的区域造影剂灌注与周围组织增强强度接近。结论 皮下及肌肉间隙内植入的丝素蛋白凝胶的降解速率基本一致,二维超声及CEUS可在体实时评估丝素蛋白凝胶的降解及血管化。 |

Objective To evaluate the feasibility of ultrasound imaging in assessing silk hydrogel biodegradation and neovascularization of silk hydrogel implanted beneath the skin or between skeletal muscles of the rat hind limbs. Methods Silk hydrogel was obtained by sonicate 8%W/V silk protein solution. Forty male Wistar rats were randomly divided into beneath skin implanted group (n=20) and skeletal muscle implanted group (n=20); 0.2 ml Cylinder-shaped silk hydrogel plugs were implanted and assessed using two-demesional ultrasound and CEUS at right time and 1, 4, 8, 12, 16, 18 weeks after implantation. The diameters and echogenicity of implants were obtained. After ultrasound imaging at each time point, two rats of each group were randomly sacrificed and the silk gel implants were excised for pathological examination. Results The implanted gel of the two groups got thinner from 12 weeks and the transverse diameter of the gels at 12 weeks were significant smaller than those after surgery right time (P<0.05). The transverse, width and longitudinal diameter of two groups at 16 weeks were significantly different from right time (all P<0.05). The contrast enhancement values obtained by CEUS showed a relatively low level (<5 dB) during 1-8 weeks, and at 12 weeks microbubbles were seen gradually penetrate into the gel area. The echogenicity increased to 20 dB at 16 weeks and >35 dB at 18 weeks and the echogenicity of gel area was similar with surrounding tissue. Conclusion The silk hydrogel biodegradation rate between skeletal muscle group and beneath skin group are similar and ultrasound imaging could noninvasively evaluate the degradation and vascularization of silk implants in vivo. |
