      目的 观察长脉冲超声(US)条件下脂质体微泡造影剂协同重组组织型纤溶酶原激活剂(rt-PA)溶解微血栓的疗效。方法 采集6名健康受试者全血制备200~300 μm的微血栓,根据不同干预方式分为单纯rt-PA组、rt-PA+低占空比US组(5000个循环)、rt-PA+高占空比US 组(600 000个循环),另将仅US(600 000个循环)照射但无声学微泡协同作为单纯US组。根据辐照系统内压力减低程度评价溶栓的疗效。结果 单纯rt-PA组溶栓率与rt-PA+低占空比US组比较,差异无统计学意义(P>0.05),rt-PA+高占空比US组溶栓率高于单纯rt-PA组和rt-PA+低占空比US组(P均<0.01)。结论 长脉冲US声学微泡辅助rt-PA溶栓效果良好,但此协同作用需在低声压条件下依赖一定比例的占空比才能实现。
英文摘要:
      Objective To observe the synergistic effect of lipid microbubble synergistic recombinant tissue plasminogen activator (rt-PA) thrombolysis under long-pulsed ultrasound (US) enhanced condition. Methods The whole blood of six healthy subjects were collected and 200—300 μm thrombi was prepared. All blood samples were divided into rt-PA group, rt-PA+low duty cycle group (5000 cycle), rt-PA+high duty cycle group (600 000 cycle) according to different intervene methods, meanwhile ultrasound irradiate without lipid microbubble as US group. Intra-system pressure was monitored to evaluate of the efficacy of thrombolysis. Results There was no significantly differences in thrombolytic rate between rt-PA+low duty cycle group and rt-PA group (P>0.05). The thrombolytic rate in rt-PA+high duty cycle group was higher than that of rt-PA group and rt-PA+low duty cycle group (both P<0.01). Conclusion rt-PA thrombolys assisted with long-pulsed US acoustic microbubbles is good, which is dependent on duty-cycle under low acoustic pressure condition.
