      目的 制备一种新型靶向微泡-干细胞复合体。方法 采用“生物素-亲和素”桥连法构建携抗ICAM-1抗体的靶向微泡(MBICAM-1)。用带正电荷的多聚赖氨酸(PLL)对MBICAM-1进行包覆和修饰,使靶向微泡表面带正电荷。采用静电吸附法将PLL修饰的MBICAM-1与大鼠骨髓间充质干细胞(MSC)共同孵育,制备MBICAM-1-MSC复合体;以流式细胞仪检测靶向微泡与MSC的结合率。结果 经PLL修饰后,MBICAM-1微泡表面带正电荷,但微泡形态、粒径以及与损伤内皮细胞(HUVEC)的结合率与未经PLL修饰的MBICAM-1无显著差异(P>0.05)。PLL修饰的MBICAM-1与MSC结合形成MBICAM-1-MSC复合体,当MSC与MBICAM-1之间的比例为1:40时,二者结合率达(29.45±2.88)%。结论 成功制备了MBICAM-1-MSC复合体;MSC与MBICAM-1的比例为1:40时,其结合率最大。

      Objective To prepare a novel targeted microbubble-stem cell complexes. Methods ICAM-1-targeted microbubbles (MBICAM-1) were prepared by using biotin-avidin bridging chemistry method. Subsequently, MBICAM-1 were coated and modified by binding positively charged polylysine (PLL) to their surface. Rat marrow mesenchymal stem cells (MSC) were mixed with PLL modified MBICAM-1, allowing for electrostatic interaction. The attachment rate of MBICAM-1 to MSCs was tested with flow cytometry. Results The surface potential of PLL modified MBICAM-1 was positive. There was no significant difference of the morphous, diameter and attachment rate to damaged human umbilical vein endothelial cells (HUVEC) between PLL modified MBICAM-1 and premodified MBICAM-1 (P>0.05). MBICAM-1-MSC complexes were prepared by successfully attaching PLL modified MBICAM-1 to MSC. Meanwhile, the attachment rate of MSC to MBICAM-1 was (29.45±2.88)% at the mix ratio of 1:40. Conclusion MBICAM-1-MSC complexes are prepared successfully using the above method. The attachment rate of MSC to MBICAM-1was maximal at the mix ratio of 1:40 (MSC/MBICAM-1).
