      目的 探讨采用99mTc-膜联蛋白Ⅴ(Annexin Ⅴ)进行静脉血栓显像的可行性。方法 将10只家兔随机分为血栓组及对照组,每组5只。血栓组通过手术于下腔静脉置入螺旋铜丝,建立静脉血栓模型;对照组手术过程与造模术相同,但不置入铜丝。术后1天对两组实验兔注射99mTc-Annexin Ⅴ进行显像,分别于注射后30、60、90 min及2 h行静态平面显像,勾画ROI,血栓组分别计算血栓部位与本底、血栓头侧下腔静脉部位放射性计数比值,对照组分别计算与血栓组血栓段对应的下腔静脉部位与本底、头侧下腔静脉部位放射性计数比值。处死动物,血栓组取血栓称重、测量放射性计数,计算每克组织百分注射剂量率(%ID/g),行HE染色。结果 血栓组5只实验兔可于血栓部位观察到放射性浓聚,对照组下腔静脉未见放射性浓聚。血栓组血栓与本底、血栓与头侧下腔静脉部位放射性计数比值(4.06±0.49、1.62±0.26)高于对照组(2.27±0.24、0.96±0.05;t=7.32、5.63,P均<0.05),血栓百分注射剂量率为 ,病理显示为新鲜混合血栓。结论 99mTc-Annexin Ⅴ有望成为一种新的静脉血栓显像剂。

      Objective To explore the feasibility of 99mTc-Annexin Ⅴ scintigraphy of venous thrombus in rabbit models. Methods Ten rabbits were randomly divided into thrombus group and control group (each n=5). Thrombus of inferior vena cava (IVC) was developed in thrombus group through surgical operation and placing spiral copper wire into IVC. The rabbits of control group received sham operation. One day after operation, planar anterior abdominal images were obtained 30, 60, 90 min and 2 h after 99mTc-Annexin Ⅴ injection. The ratios of thrombus to background and thrombus to head lateral IVC were calculated by the counts of ROI. Percentages of the injected dose per gram of ex tissue of thrombus were calculated. Paraffin slices of thrombus were stained by HE stains. Results 99mTc-Annexin Ⅴ uptake in thrombi were clearly visualized in all rabbits of thrombus group. All rabbits of control group showed negative images. The thrombus to background and head lateral inferior vena cava ratios (4.06±0.49, 1.62±0.26) of the thrombus group were higher than those of control group (2.27±0.24, 0.96±0.05;t=7.32, 5.63;all P<0.05). The percentage of injected dose per gram of thrombi was (0.0189±0.0022)%ID/g in thrombus group. All thrombi of thrombus group were confirmed fresh mixed thrombi. Conclusion 99mTc-Annexin Ⅴ may become a new acute venous thrombus imaging tracer.
