      目的 利用声脉冲辐射力成像(ARFI)技术定量观察谷氨酰胺对大鼠非酒精性脂肪性肝病(NAFLD)的治疗作用。方法 将36只雄性SD大鼠随机均分为对照组、模型组和谷氨酰胺治疗组,检测各组大鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、甘油三酯(TG)和胆固醇(TC)水平,测量肝组织谷胱甘肽(GSH)、肿瘤坏死因子α(TNF-α)和丙二醛(MDA)水平;观察肝脏组织病理改变;应用ARFI获取肝脏不同区域剪切波速度(SWV)。比较不同组别大鼠上述指标的差异。结果 建模后12周末模型组血清ALT、AST、TG和TC,肝组织TNF-α和MDA水平明显高于对照组,GSH水平明显低于对照组(P均<0.05),谷氨酰胺治疗后上述指标均有所改善(P均<0.05);8周末和12周末高脂模型组肝前区和肝中区SWV值均较对照组明显增加(P均<0.05),谷氨酰胺治疗组较高脂模型组有所降低(P<0.05)。结论 谷氨酰胺对大鼠NAFLD具有一定治疗作用;ARFI可对其进行定量评估。

      Objective To quantitatively assess the impact of glutamine on nonalcoholic fatty liver disease (NFLD) in rats with acoustic radiation force impulse (ARFI). Methods Totally 36 SD rats were randomly divided into control group, model group and glutamine treated group (each n=12). The levels of alanine transaminase (ALT), aspartate aminotransferase (AST), triglyceride (TG) and total cholesterol (TC) in serum were measured, of glutathiose (GSH), tumor necrosis factor-α (TNF-α) and malonaldehyde (MDA) in the liver were detected. Histopathologic changes of the liver were observed under microscopy. The shear wave velocity (SWV) in different regions of the liver was obtained in ARFI. All above indexes were compared among 3 groups. Results The levels of ALT, AST, TG, TC, TNF-α and MDA in model group were significantly higher and GSH was lower than those in control group (all P<0.05), which were all significantly improved by glutamine (all P<0.05) on the 12th week after establishment of models. SWVs of anterior and middle region of liver in model group significantly increased than those in control group (both P<0.05), which obviously decreased after glutamine administration (both P<0.05) on the 8th and 12th week. Conclusion Glutamine has certain therapeutic effect on NAFLD in rats, which can be observed quantitatively with ARFI.
