       目的 探索高频超声引导注射技术实现主要组织相容性(抗原)复合物(MHC)基因大鼠胸腺转移的新方法。方法 应用分子生物学技术获得供体Balb/c小鼠异种MHC-I类抗原(H-2Kd)基因,并构建pCI-neo-H-2Kd质粒。对20只成年SD大鼠应用高分辨力超声影像系统获得胸腺标准切面超声声像图,同时使用Vevo轨道系统和超声引导注射针装置,在超声引导下将质粒注入大鼠胸腺内。应用流式细胞仪检测H-2Kd基因在转染大鼠胸腺细胞上的表达。结果 超声引导下注射质粒后全部大鼠存活。H-2Kd基因成功转入受体大鼠胸腺,流式细胞仪检测H-2Kd在大鼠胸腺内的表达阳性率为60.73%(56.04%~62.49%)。结论 高频超声引导注射技术定位准确,方法简便,对组织损伤轻微,注射成功率较高,可代替手术胸腺内注射法诱导免疫耐受。

      Objective To explore the feasibility of image-guided intrathymic injection of major histocompatibility complex (MHC) using high-resolution ultrasound. Methods The xenograft MHC-I antigen (H-2Kd) gene was extracted and proliferated from donor Balb/c mice, and the expression vector plasmid of pCI-neo-H-2Kd was constructed with molecular cloning technique. Images on standard section were obtained with high-resolution imaging system. Then the plasmid of pCI-neo-H-2Kd was injected into the thymus of 20 Sprague-Dawley rats with Vevo rail system and ultrasound-guided fine-needle. After injection, expression of H-2Kd in the thymocytes was tested by fluorescence-activated cells sorter (FACS). Results Injection was successful in all the rats. H-2Kd gene was expressed in the receptors' thymus with a transfection efficiency of 60.73% (56.04%-62.49%). Conclusion Ultrasound-guided intrathymic injection is a safe, effective and convenient way with high degree of target accuracy to perform gene transfection to thymus. This method might induce immune tolerance and instead of open-chest intrathymic injection.
