目的 制备鸟嘌呤(G)含量及离启动子远近不同的4种脂质体包裹99mTc-HYNIC-Survivin ASODN对荷人肝癌裸鼠行SPECT显像,探讨其显像质量是否受G含量及启动子位置影响,以找到最佳反义显像的碱基序列,提高基因显像质量。方法 制备两种(A1、A2)G含量不同而离启动子远近相近的16个碱基单链ASODN和两种(A'1、A'2)G含量相同而离启动子远近不同的10个碱基单链ASODN,耦联HYNIC,99mTc标记后,经Cellufine GH-25纯化后脂质体包裹。用此显像剂对荷瘤裸鼠行肿瘤反义显像并通过ROI技术进行分析。结果 肿瘤/对侧肌肉比值(T/NT)A1、A2分别为4.13±0.09 和2.47±0.04(2 h),4.90±0.11和3.75±0.10(4 h);A'1、A'2分别为3.90±0.07和2.09±0.07(2 h),4.41±0.08和2.78±0.10(4 h)。上述各组T/NT值在各时间点差异有统计学意义(P<0.01)。结论 G含量相同,离启动子位置较近,显像质量较好;离启动子位置相近,G含量较低,显像质量较好。 |

Objective To find whether imaging quality of 99mTc-HYNIC-Survivin ASODN in nude mice bearing human hepatocellular carcinoma was influenced by their guanine content and promoter's position using four different kinds of synthesized and liposome-coated 99mTc-HYNIC-Survivin ASODN, and to find optimal antisense imaging sequence for improving the quality of genic imaging. Methods Two kinds (A1, A2) of synthesized Survivin ASODN with two guanine contents (12.5%, 50.0%) and close promoter's position, 16 bases per single-stranded and two kinds (A'1, A'2) of synthesized Survivin ASODN with same guanine content and different promoter's position, 10 bases per single-stranded were prepared and labeled with 99mTc by conjugating HYNIC, purified through Cellufine GH-25, and then encapsulated with cationic. Antisense gene imaging of 99mTc-HYNIC-Survivin ASODN in nude mice bearing tumor were drew and analyzed with ROI technique. Results The ratio of tumor to muscle (T/NT) was 4.13±0.09 and 2.47±0.04 at 2 h after intravenous injection of 99mTc-HYNIC-Survivin ASODN (A1, A2), and 4.90±0.11 and 3.75±0.10 at 4 h in antisense gene imaging study, respectively. The ratio was 3.90±0.07 and 2.09±0.07 at 2 h after intravenous injection of 99mTc-HYNIC-Survivin ASON (A'1, A'2), and was 4.41±0.08 and 2.78±0.10 at 4 h in antisense gene imaging study, respectively. T/NT value of all groups was significant difference at different time points (P<0.01). Conclusion The antisense gene imaging was better in if guanine contents was same and the ASODN sequence was closer to the promoter's position, or the distance between ASODN and promoter's position was close and guanine content was lower. These results may provide some guidance for improving clinical tumor antisense imaging. |
