      目的 利用组织多普勒成像(TDI)技术评价从幼年到老年不同月龄的正常Wistar大鼠心脏舒张功能的增龄性改变。方法 选用月龄分为1、2、5、12、20个月的正常雄性Wistar大鼠60只,分别行经胸超声心动图检查,最后处死动物取左心室称重并送组织病理学检测。结果 随增龄各组大鼠左心室相对质量(LVRM)明显增加(P<0.01),于20个月增至最重。超声所计算的LVRM与实体标本所测值呈正相关(r=0.780, P<0.001)。二尖瓣口舒张末期血流速度(E)20月龄组与各低龄组相比较组间无统计学差异(P>0.05),二尖瓣环舒张早期最大运动速度(Em)随月龄增长逐渐下降,二尖瓣环舒张晚期最大运动速度(Am)峰值呈上升趋势,Em/Am比值随月龄增长逐渐下降,Em/Am比值在1~5个月龄组>1,12个月龄和20个月龄组<1。组织病理学检测:20个月龄组心肌出现灶性纤维化。结论 DTI技术能够检测大鼠左心室舒张功能和观察其增龄性改变的规律,为基础实验研究提供可靠的无创方法及依据。
英文摘要:
      Objective To evaluate the age-related changes in left ventricular diastolic function on various age Wistar rats with tissue doppler imaging (TDI). Methods Sixty male Wistar rats divided into 5 groups (aged 1, 2, 5, 12 and 20 months were enlisted, each group n=12). All groups were examined in echocardiography. Finally, histopathologic examination was performed. Results Left ventricular myocardial weight of rats increased with aging (P<0.01), and the heaviest was in 20 months of age. There was positive correlation between left ventricular mass examined by echocardiography with that examined by necropsy (r=0.803, P<0.001); the maximal early diastolic velocities (Em) of mitral annulus and Em/Am ratio were significantly decreased by aging (P<0.01), however, the maximal later diastolic velocities (Am) of mitral annulus has a tendency to increase with aging. Em/Am ratio>1 at 1, 2, and 5 months group, conversely Em/Am ratio<1 at 12 and 20 months group. Histopathologic study demonstrated focal myocardial fibrosis in 20 months group. Conclusion TDI is a reliable non-invasive method which can substitute the invasive hemodynamic method to evaluate age-associated changes in rats left ventricular diastolic function.
