目的 探讨和优化纤维蛋白靶向对比剂(EP-2104R)对兔颈动脉急性和亚急性血栓进行3.0T MR的检查序列和检查时间。 方法 以颈动脉外部损伤和阻塞法制备兔颈动脉内血栓形成模型,分为急性期组(6 h)和亚急性期(6 d)组。分别于EP-2104R注入前及注入后不同时间点行MR黑血技术扫描,观察血栓形态及信号等特点,计算血栓对比噪声比(CNR),并与组织病理学对比。 结果 在T1WI双翻转快速自旋回波压脂序列(T1WI-DIR压脂序列),EP-2104R准确地显示所有颈动脉血栓,呈高信号亮点,信号均匀,边缘清楚,优于非对比增强MRI (P<0.01)。亚急性期血栓的CNR明显高于急性期血栓的CNR (P<0.001)。血栓CNR在2~3 h达到高峰。 结论 在兔颈动脉血栓形成模型,于T1WI-DIR压脂序列,EP-2104R可以准确显示活体内急性期及亚急性期血栓,优于非对比增强MRI;对活体内不同时期血栓的理想检查时间窗为注射EP-2104R后2~3 h。 |

Objective To optimize the 3.0T MRI sequences and examining time, and to evaluate the use of Fibrin-targeted contrast agent (EP-2104R) for MRI thrombus detection at acute and subacute period after thrombus generation in rabbit models. Methods The carotid artery thrombosis models were established with external injury and stasis and were divided into acute phase thrombosis group (6 h) and subacute phase thrombosis group (6 d). MR black blood technology was performed before and after EP-2104R injection. The patterns and signal characteristics of thrombosis were observed, contrast-to-noise ratio (CNR) was calculated, and then correlation between images and histopathology was compared. Results By T1-weighted double-inversion recovery, fast spin echo sequences and fat-suppressed sequence (T1WI-DIR FS), EP-2104R accurately showed all the carotid artery thrombosis with high, even signal and clear edge, superior to non-contrast-enhanced (P<0.01). The CNR of the subacute phase thrombosis was significantly higher than that of the acute phase thrombosis (P<0.001). The peak of CNR of thrombosis was at 2-3 h. Conclusion In the rabbit carotid artery thrombosis model, using T1WI-DIR FS sequence, EP-2104R can accurately show the acute phase and subacute phase thrombosis in vivo, being superior to non-contrast-enhanced MRI. The ideal examining time window is 2-3 h after EP-2104R injection. |
