目的 观察单次大剂量碘对大鼠甲状腺内钠碘转运体(NIS)蛋白表达量的影响,探讨应用99mTcO-4进行甲状腺扫描前是否应对碘的摄入进行限制。方法 将SD大鼠分成给碘后1 d处死组、给碘后7 d处死组和对照组,每组8只;将2000 μg碘化钠(NaI)溶于0.5 ml生理盐水注入前两组大鼠腹腔内,对照组注射等量生理盐水。各组大鼠处死后,取其甲状腺,应用Western免疫印迹方法探测NIS蛋白含量,通过光密度分析方法对信号强度进行半定量分析。结果 给碘后1 d处死组大鼠甲状腺内NIS蛋白含量明显低于对照组和给碘后7 d处死组(P均<0.001), 而给碘后7 d处死组与对照组间NIS蛋白表达量的差异无统计学意义(P>0.05)。结论 单次大剂量碘摄入可降低甲状腺内NIS蛋白的表达量,此效应在给碘1 d后明显,7 d后可恢复至正常水平。 |

Objective To investigate if there needs limitation to iodide intake before thyroid scintigraphy with 99mTcO-4 by evaluating effect of a single dose of iodide intake on rat thyroid sodium/iodide symporter (NIS) protein expression. Methods Sprague Dawley (SD) rats were divided into 1 day group, 7 day group and control group with 8 animals in each group. The animals in 1 day group and 7 day group were administered 2000 g NaI dissolved in normal saline through peritoneal injection, and animals in control group were given equal dose of normal saline. The animals in 1 day group and 7 day group were sacrificed on day 1 and day 7 after iodide was administrated, and thyroid was isolated to detect the NIS protein expression using Western blot and semi-quantitative analysis was done using optical density analysis. Results The NIS protein expressed in 1 day group was significantly lower than its expression level in 7 day group and control group (both P<0.001), while there were no significant difference between control group and 7 day group (P>0.05). Conclusion A single large dose of iodide intake can significantly decrease thyroid expression of NIS protein on 1 day after iodide was administrated, and expression of NIS protein can return to normal level 7 days later. |
