      目的 制作一种与生物组织热凝固温度相似的可视化仿组织体模并应用于射频消融研究。 方法 仿组织体模主要由作为温度显示剂的牛血清白蛋白(BSA)和聚丙烯酰胺凝胶(PAG)构成,制作过程中通过柠檬酸缓冲液将其pH调节至4.3~4.7范围内。测定体模的透光度、吸光率、密度、导电率和比热等参数,并用射频对体模进行消融测试。 结果 新制的体模为淡琥珀色、透明、韧性好、脆性小的固体状胶体。室温25℃时,体模在热凝固前的透光度为95%,吸光率为0.021,而热凝固后其透光度小于10%,吸光率为1。体模(pH=4.3)的密度为1.070 kg/m3,在温度37℃及频率为470 kHz下的导电率为0.116 s/m,比热为3567 J/(kg·℃)。加热后凝固灶呈乳白色,边界清楚。 结论 pH调节后的可视化PAG仿组织体模的凝固温度可下降至与生物组织热凝固温度相同范围,体模具有与生物组织相似的物理特性,适合于RFA的实验研究。

      Objective To develop a tissue-mimicking phantom for visualizing three-dimensional coagulation temperature distribution at the same range of biological tissue and to visualize the coagulation lesion of radiofrequency ablation (RFA) using phantom. Methods The pH of the phantom, which mainly contained polyacrylamide gel (PAG) and bovine serum albumin (BSA) as a temperature sensitive indicator, was adjusted to the range of 4.3-4.7 using the citrate buffer. The parameters of phantom, including density, transparence, absorbance, electrical conductivity, and specific heat capacity, were measured. In addition, the phantom was investigated using radiofrequency ablation. Results The new phantom is a gel in solid form with light amber, optical transparency and sufficient strength with lower fragility. Before thermal ablation, the transparence and absorbance of phantom were 95% and 0.021 at 25℃, respectively. However, after thermal ablation, the transparence of phantom was less than 10% and the absorbance was 1. The density, electrical conductivity and specific heat capacity of phantom (pH=4.3) were 1.070 kg/m3, 0.116 s/m (37℃, 470 kHz), and 3567 J/(kg·℃), respectively and similar to those of the human liver. The coagulation lesions are ivory white and have a distinct boundary with the uncoagulation regions. Conclusion The phantom, whose pH was adjusted by buffering, can visualize three-dimensional temperature distribution at the same range of coagulation temperatures observed in tissue during thermal ablation. The physical properties of the phantom are favorable for RFA study.
