      目的 研究阿霉素海藻酸钠微球对兔VX2肝移植瘤的化疗栓塞作用。方法 30只新西兰大白兔,于肝左叶内植入VX2 肿瘤组织块,建立肝移植瘤模型。实验动物随机分成5组,每组6只。各组实验动物均开腹游离肝动脉,经肝动脉楔入法分别给予生理盐水(A组)、空白海藻酸钠微球(B组)、阿霉素海藻酸钠微球(C组)、超液化碘油(D 组)、超液化碘油+阿霉素(E 组)。治疗后第3周将所有实验动物处死,取肿瘤标本进行病理组织学检查,测定不同处理组肿瘤生长率、坏死率、凋亡指数,并观察不同处理组肝内转移及远隔转移的影响。结果 阿霉素海藻酸钠微球组明显抑制肿瘤生长,移植瘤坏死率高于空白微球组;在肝内及远隔转移方面,空白微球组及载药微球组抑制远隔转移方面较其他组优,两组间相比则无显著性差异(P>0.05)。阿霉素海藻酸钠微球组TUNEL阳性率最高,为(14.3±3.65)%。结论 阿霉素海藻酸钠微球通过栓塞肿瘤供血动脉,局部缓慢释放化疗药物,可以提高兔VX2 肝移植瘤模型的化疗效果。

      Objective To evaluate the antitumor effect of transcatheter arterial chemoembolization (TACE) with alginate microsphere-adriamycin in experimentally induced liver tumor. Methods Thrity New Zealand white rabbits were randomly divided into five groups and VX2 carcinoma was grown in the left lobes of the livers.TACE was performed with normal saline (Group A), alginate microsphere (Group B), alginate microsphere-adriamycin (Group C), Lipiodol (Group D), and Lipiodol-adriamycin (Group E). Three weeks later, the animals were killed and pathologic parameters were calculated on the basis of findings. Effects on intrahepatic and distal metastasis in all groups were examinated. Results Alginate microsphere-adriamycin suppress the growth of tumor and have a high level of necrosis.In the groups of using Alginate microspheres,there were less intrahepatic and distal metastasis comparing with the others.But no difference can be seen between Group B and Group C. High level of positive cells in TUNEL (14.3%±3.65%) were observed in Group C. Conclusion Alginate microspheres can potentially serve as embolizing agents and drug delivery vehicles for local slow-release. Chemoembolization with Alginate microsphere-ADM is an effective antitumor treatment of hepatic carcinoma in an experimental model.
