目的 研究年龄在E-选择素G98T基因多态性对高血压影响中的作用。方法 高血压组326例,对照组216例。对两组进行血压测量,检测生化指标及NO、ET、AI、AII、ALD。以PCR-RFLP方法进行DNA多态性分析,电泳判断基因型并测序。结果 检测到TT、GG、GT三种基因型。在<40岁组,携带T等位基因发生高血压的危险性为G的6.294倍(P=0.007;OR=6.294;95%CI:1.361~29.11 )。在40~60岁及>60岁组,高血压组与对照组的等位基因频率则无显著差异(P=0.528和0.965)。<40岁的HT组中TT-TG基因型CSBP、CDBP及MAP显著高于GG型(P分别为0.006、0.023和0.036)。在40~60岁组:TT-TG基因型的CSBP、MAP也显著高于GG基因型(P分别为0.031和0.042),而>60岁组,两基因型间血压无显著差异。在<40岁的HT组中TT-TG基因型LVMI显著大于GG型(P=0.042)。结论 E-选择素G98T基因多态性对高血压有显著影响,T等位基因携带者发生高血压风险大,血压值较高,更倾向于发生左室肥厚。这种影响随着年龄的增长而减弱。 |

Objective To investigate how age affect the effect of E-selectin G98T polymorphism on blood pressure. Methods Genotypes of 326 essential hypertension patients (male 173, female 153, age y, BMI kg/m2) and 216 healthy individuals (male 60,female 156, age y, BMI kg/m2) were typed by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP). Took blood pressure and examed BG, TG, TC, LDL-c, HDL-c, UA, BUN, NO, ET, AI, AII, ALD of each individuals. Results Three genotypes (TT, GG, GT) were found. In <40 y group, risk of hypertension for allele T is 6.294 times of that for allele G (P=0.007; OR=6.294; 95%CI: 1.361-29.11). While in 40-60 y and >60 y group, there were no significantly different in genotype between HT group and N group. In <40 y HT group, the CSBP, CDBP and MAP of TT-TG genotype are higher than that of GG (P=0.006, 0.023 and 0.036). In the same group, the LVMI of TT-TG is higher than that of GG (P=0.042). Conclusion E-selectin G98T polymorphism affect blood pressure significantly. T allele has higher risk for hypertension and left ventricular hypertrophy. But this effect are affected by age. |
