      目的 研究兔VX2肝移植瘤的微血管密度(MVD)和血管内皮生长因子(VEGF)与CT灌注成像各灌注参数的相关关系。方法 对16只VX2肝移植瘤模型兔(种植后2周)行CT灌注扫描,获得各灌注参数值,对肿瘤周边CT强化较明显区域行VEGF、CD34的免疫组化染色,根据CD34阳性结果计数测定MVD,分析灌注参数与MVD、VEGF的相关性。结果 瘤灶中心及边缘区MVD、VEGF阳性表达有明显差异(P<0.01),瘤灶边缘区MVD值随着VEGF表达增强而增高,瘤灶边缘区各灌注参数值:肝总血流灌注量(HBF)、毛细血管表面渗透性(PS)、肝动脉灌注指数(HAI)、肝动脉灌注量(HAP)与MVD、VEGF 呈正相关(P<0.01),门静脉灌注量(HPP)与MVD、VEGF 呈负相关(P<0.01),平均通过时间(MTT),肝血容积(HBV)与MVD、VEGF 无相关性(P>0.05)。结论 多层螺旋CT灌注参数与MVD、VEGF表达有明显相关性,可用于活体评价肿瘤血管生成。
英文摘要:
      Objective To study the relationship of CT perfusion parameters with microvessel density (MVD) and vascular endothelial growth factor (VEGF) in rabbit VX2 liver tumor. Methods Sixteen rabbit-models underwent CT perfusion, and the perfusion parameters were analyzed. The enhanced border of tumors were stained immunohistochemically with VEGF and CD34, MVD was shown by anti-CD34. The correlation of perfusion parameters with MVD and VEGF were analyzed. Results The positive expressions were different significantly in center and border of the tumor (P<0.01). There was significant correlation between MVD and VEGF in the border of the tumor, the more VEGF expressed, the more MVD increased. The correlation analysis were done between perfusion parameters and MVD, VEGF in the border of the tumor. Positive relations were shown between the value of HBF, PS, HAI, HAP and MVD, VEGF, and negative relations were shown between HPP and MVD, VEGF, but no relations between the value of MTT, HBV and MVD, VEGF. Conclusion There was significant correlation between perfusion parameters and MVD, VEGF. The perfusion parameters can be used to evaluate tumor angiogenesis in vivo.
