目的 探讨骨髓干细胞移植于缺血心肌后,应用心肌声学造影技术检测其微血管新生效应的价值。方法 18只兔随机分3组:细胞移植组(n=8,心梗后移植)、心肌梗死组(n=6,心梗后不治疗)、对照组(n=4,无心梗不治疗)。经静脉注射声学造影剂,检测各组动物心肌梗死区移植前后造影剂充盈状况、造影剂峰值强度。取心肌组织测定新生毛细血管密度。结果 移植4周后,移植组缺血心肌内可见少量造影剂回声,心梗组未见造影剂回声,对照组造影剂充盈良好。造影剂峰值强度在各组间差异显著(P<0.01)。心肌内造影剂的声强度与病理检测的毛细血管密度存在正相关关系(r=0.77)。结论 心肌声学造影技术能有效检测干细胞移植后的微血管新生效应,可作为临床上评价和随访细胞移植疗效的方法。 |

Objective To certificate the clinic value of using myocardial contrast echocardiography (MCE) to detect the neovascularization effects of bone marrow stem cells transplantation into ischemic myocardium. Methods Eighteen rabbits were divided into 3 groups randomly as follow: group A (n=8, with acute left ventricular anterior myocardial infarction, and accept autologous bone marrow stem cells implantation), group B (n=6, with AMI, without treatment but medium injection) and group C (n=4, without AMI and treatment). MCE was executed on all animals through inputting contrast agent intravenously. The microbubbles infilling level and intensity of ischemic area were detected before and after transplantation. Microvessels density was determined pathologically. Results Microbubbles was distributed homogenously in the left ventricular anterior wall of group C, and was defected obviously in group B, while contrast agent echo could be observed in group A. Compared with group B, contrast agent peak intensity was significantly increased in group A(P<0.01). There was a positive correlation between the microvessles density pathologically and the microbubbles intensity echocardiographically (r=0.77). Conclusion MCE could detect the neovascularization caused by bone marrow stem cells transplantation into ischemic myocardium with efficiency and could be regarded as a way to assess and follow-up stem cells transplantation effects clinically. |
