目的 通过对兔脑VX2种植瘤放疗前后CT灌注参数与肿瘤微血管密度(MVD)的相关性研究,为CT灌注成像成为脑转移瘤放疗疗效评估新的影像学手段提供实验依据。方法 兔脑VX2种植瘤模型9只,分为治疗组4只,非治疗组5只,治疗组做肿瘤放疗。两组均于放疗前、后一周行CT灌注成像及增强扫描。处死后行脑组织HE及CD34免疫组织化学染色。分别对治疗组与非治疗组放疗前后的CT灌注参数进行显著性分析及放疗后两组肿瘤区的CT灌注参数值CBV值与肿瘤MVD作相关性分析。结果 治疗组放疗后瘤区及瘤周区CT灌注参数CBF值、CBV值、PS值均较放疗前降低,P<0.05,有统计学差异;非治疗组前后两次CT灌注成像检查瘤区、瘤周区CT灌注各参数值均无明显改变,P>0.05,无统计学意义。肿瘤实质区的CBV值与CD34免疫组化MVD计数值间具有显著的正相关(r=0.896,P<0.01)。结论 CT灌注图像能反映VX2种植瘤放疗前后血流动力学改变,瘤区CBV值与组织学MVD值间具有显著的相关性,可作为一种活体评价肿瘤微血管的指标。 |

Objective With correlative study of CT perfusion parameter change and tumor micro-vascular density change of rat VX2 implanted cerebral tumor before and after radiation therapy, this study was supposed to provide experimental basis for evaluation of CT perfusion imaging in measurement of effect assessment of radiation therapy of brain tumor. Methods Nine rats with VX2 tumor were involved, and all rats were divided into treatment group (n=4) and non-treatment group (n=5). The rats in treatment group underwent brain directional radiation therapy. CT enhanced and perfusion scans were performed one week both before and after the radiation for the two groups. Brain tissue was collected to perform HE stain and CD34 immuno-histo-chemical stain after all rats were sacrificed to death. Significant analysis of CT perfusion parameters was applied between before and after the radiation therapy of treatment and non-treatment group respectively. Correlation study was also made between CBV value and MVD value of the tumor region in treatment group after radiation therapy and in the non-treatment group as well. Results All the CT perfusion parameters including CBF, CBV and PS decreased after radiation in treatment group with statistic significance (P<0.05). All the CT perfusion parameters had no obvious decrease twice in non-treatment group(P>0.05). Significant correlation was observed between CBV and MVD value after CD34 immuno-histo-chemical stain in tumor region(r=0.896,P<0.01). Conclusion CT perfusion imaging is able to reflect hemodynamic changes of brain tumors after radiation therapy. Cerebral blood volume value can be taken as an index to the micro-vascular of tumor in-vivo as it has significant correlation with microvascular density. |
