      目的 对比研究诊断用彩色多普勒超声以不同时限单次及重复照射对胎鼠大脑皮层神经元P53蛋白表达及超微结构的影响。方法 彩超照射孕16天SD大鼠子宫体表投影区,按照射次数分为单次照射和重复照射两大组,重复照射组为首次照射间隔48 h后,相同声场条件下再照射一次;以上两大组再按每次照射时限的不同各自划分为照射10 min组、20 min组、30 min组及对照组。各组于既定时间照射后24 h取胎鼠脑组织标本,免疫组化方法检测P53蛋白的表达,透射电镜观察超微结构的变化。结果 照射10 min时:单次照射组、重复照射组P53蛋白表达与对照组无显著差异(P>0.05)超微结构亦未见明显异常。照射20 min、30 min时:单次照射组及重复照射组P53蛋白表达均增强,与对照组有显著差异(P<0.05);且重复照射组与单次照射组之间也有显著性差异(P<0.05)。电镜观察见神经元染色质浓缩边集、线粒体肿胀变性及神经元突起肿胀变性,重复照射30 min组表现最为明显且伴有血管内皮细胞的超微结构改变。结论 ①晚孕期彩超照射SD大鼠子宫投影区超过20 min,胎鼠大脑皮层神经元p53表达增加,超微结构出现改变;②相同照射时限内重复照射对胎鼠脑组织p53表达及超微结构的改变明显高于单次照射。

      Objective To study the changes of protein P53 expression and ultrastructure of fetal rats insonificated by repeated color Doppler ultrasound (CDUS). Methods Pregnant 16 days SD rats were insonificated by diagnostic CDUS, and they were divided averagely and randomly according to insonification time (10 min, 20 min, 30 min) and whether the repeated insonification was adopted or not, Control group was sham-insonificated 24 hours later. The fetal rats' brains were resected for further research. P53 expression was examined by immunohistochemistry method,and utrastructural changes were observed by transmission electron microscope (TEM). Results There were no significant changes on expression of protein P53 and ultrastructural changes in 10 min and repeated 10 min groups, but there were significant changes in 20 min and 30 min groups, especially repeated groups. And there were significant differences between single insonification 20 min and 30 min groups and repeated insonification 20 min and 30 min groups (P<0.05). Some ultrastructural changes in neuron cells such as chromatin concentration/marginztion, mitochondria swelling were observed by TEM test. Conclusion Repeated insonification with diagnostic CDUS may affect the expression of protein P53 and bring changes of ultrastructural in fetal rats' central nervous system.
