      目的 探讨彩色多普勒超声照射对中孕期胎鼠肾小管上皮细胞bcl-2蛋白表达的影响。方法 32只孕14天 SD大鼠按彩超辐照时间不同随机等分为四组,Ⅰ组(对照组)、Ⅱ组(照射10 min组)、Ⅲ组(照射20 min组)、Ⅳ组(照射30 min组),仪器采用Acuson公司Sequoia-512型彩色电脑声像仪,照射条件:4V1探头,二维频率3.0 MHz,彩色频率 3.0 MHz,Tis =1.8,MI =1.6。照射后24 h 剖宫取胎鼠肾脏标本,HE染色光镜观察组织结构;免疫组化ABC法检测肾脏bcl-2蛋白的表达,采用Leica Q 500 MC型图像分析仪进行图像分析。结果 Ⅰ~Ⅳ组光镜下观察肾脏组织结构均未见异常;免疫组化法检测,各组胎鼠肾小管上皮内可见均匀散在的棕黄色淡染的bcl-2蛋白阳性表达,图像分析统计结果显示:Ⅱ组及Ⅲ组胎鼠肾小管上皮bcl-2蛋白阳性表达强度与Ⅰ组无显著差异(P>0.05);Ⅳ组表达强度与Ⅰ组有显著差异(P<0.05)。结论 彩色多普勒超声照射中孕期大鼠超过30 min可引起胎鼠肾小管上皮细胞bcl-2蛋白表达减少。

      Objective To study the effect of diagnostic CDFI on the expression of bcl-2 protein in fetal rat's renal tubule epithelial cell. Methods Thirty-two female Spraque-Danles rats that have been pregnant for 14 days were divided into four groups averagely and randomly, control group (groupⅠ) was sham-irradiation, groupⅡ-Ⅳ were irradiated by diagnostic CDFI (Sequoia-512, 4V1, 3.0 MHz, Tis=1.8, MI =1.6) for 10 min, 20 min and 30 min respectively, fetal rats' kidneys of four groups were removed at 24 h after irradiation, HE stain was done for microscope observing, expression of bcl-2 protein was detected with immunohistochemical method. Results There was no abnormal changes among group Ⅰ-Ⅳ under microscope, and there was no significant differences in expression of bcl-2 protein among group Ⅰ,group Ⅱ and group Ⅲ (P>0.05). But there was significant difference in expression of bcl-2 protein between groupⅠand group Ⅳ (P<0.05). Conclusion Continuous irradiation beyond 30 minutes by diagnostic CDFI may result in decreasing expression of bcl-2 protein in fetal rat's renal tubule epithelial cell.
