| 目的 探讨复方中药99克星经皮瘤内注射诱导肝细胞癌凋亡的生物效应。方法 取人肝癌SMMC-7721裸鼠皮下移植与肝内原位移植模型各14只,随机分为99克星治疗组14只,无水乙醇与生理盐水治疗组各7只,每隔5天分别向肿瘤内注入各组治疗药物共4次,20天后处死,取肿瘤中心组织行电镜检查与FCM DNA分析。所有实验均在双盲条件下进行。结果 盐水组治疗后肝癌细胞超微结构无明显改变;乙醇组出现以凝固性坏死为主的变化;克星组有92.9%(13/14)见到凋亡细胞或/和凋亡小体。FCM DNA分析结果克星组竟有高达92.9%(13/14)出现肿瘤程序性坏死的凋亡峰,而盐水组却有57.1%(4/7)出现异倍体峰。结论 复方中药99克星具有较强的诱导与促进细胞自发凋亡的作用,似有唤起"自杀基因"的功能,值得深入探讨。 |

| Objective To probe the biological effect of percutaneous intratumoral injection of Chinese traditional medicine Star-99 in inducing the apoptosis of the hepatic cancer cell. Methods Twenty-eight human hepatic carcinoma SMMC-7721 transplantation nude mice, of which 14 were hypodermic implanted patterns and 14 were liver orthotopic transplanted patterns, were randomly divided into three groups, which were Star-99 group containing 14 patterns, alcohol group and saline group containing 7 patterns respectively. The medicine were injected into the tumors of all the nude mice every 5 days. The nude mice were killed after being injected 4 times, 20 days after the first injection.The center tissures of the tumors were examined by electronic microscope and analysed by FCM DNA. Double blind was applied in the experiement. Results The ultrastructures of the hepatic cancer cells of the saline group after treatment were not remarkably changed. The major pathological change in the alcohol group was coagulation. The changes in the Star-99 group were multiple. It was worth noticing that the apoptosis cells or/and bodies could be seen in 92.9 % (13/14) of the tumors in the Star-99 group. The FCM DNA analysis showed that 92.9 % (13/14) in the Star-99 group appeared the apoptosis peaks of the tumor program necrosis. Yet 57.1%(4/7) in the saline group appeared the heteroploid peaks. Conclusion The compound Chinese medicine Star-99 has stronger effects of inducing and promoting the cellular self-apoptosis, seem to have the function of arousing "suicide gene" and worth deeply studying. |
