目的 研究乏氧显像剂99mTc-HL91 作为肿瘤阳性显像剂,以及肿瘤坏死对其显像的影响。方法 从人乳腺癌荷瘤裸鼠尾静脉注射99mTc-HL91 74 MBq,采集荷瘤裸鼠图像,处死后测量各脏器、组织和肿瘤的重量和放射性,肿瘤切片,进行放射性自显影和显像。结果 肿瘤显影清晰,注射后6 小时显像上,肿瘤和头部、胸部的放射性比值分别为2.44 ±0 .53 、2 .13 ±0 .64 .非坏死肿瘤和坏死肿瘤的放射性比值为3.77 ±0 .35 .肿瘤切片的放射性自显影和显像上可见坏死部分放射性低于非坏死部分。肿瘤和腹部、肝脏和肠的放射性比值低。结论 人乳腺癌荷瘤裸鼠的瘤体在99mTc-HL91 显像上为清晰的阳性病灶;坏死肿瘤组织的放射性摄取低于非坏死肿瘤;99mTc-HL91在腹部脏器滞留量高,影响其在腹部肿瘤诊断中的应用。 |

Objective To evaluate the hypoxia avid agent 99mTc-HL91 as the tracer of tumor “hot spot image” and the influence of tumor necrosis on the image.Methods Six nude mice bearing human breast cancer MCF 7 were studied. ① Gamma camera imaging:Injection of 37 MBq(1 mCi)of 99mTc-HL91 was followed by static planar imaging.② Postmortem analysis:Mice were sacrificed after injection of 99mTc-HL91 6 hour, the organs and tumor were removed, weighted, measured with radioactivity. ③ Autoradiograph and image of tumor.Results ① Gamma camera imaging:The image of tumor was identified after injection of 99mTc-HL91 1 hour. Images demonstrated increased 99mTc-HL91 uptake in the tumor after injection 1 to 6 hours, gradually(P <0.05~0.001). ② Postmortem analysis:The radioactivity ratios of tumor to brain, muscle, blood, heart and lung were higher than 4.9 after injection 6 hour. The radioactivity of non necrotic tumor was 3.77 times of that of necrotic tumor. However, the radioactivity ratios between tumor to liver, intestine and stomach were low. ③ The autoradiograph and image of tumor showed obviously higher radioactivity in the region of solid tumor than that in the necrotic region of tumor.Conclusion 99mTc-HL91 positively identifies regional tumor in nude mice bearing breast cancer MCF 7 using gamma camera. Necrotic tumor has significantly lower 99mTc-HL91 retention than non necrotic tumor does. The low radioactivity of tumor to organs in abdomen is the limitation of application of 99mTc-HL91 in detecting tumor in abdomen. |
